RESUMO

Case description: A case of a 37-year-old female patient suffering from refractory bilateral trigeminal neuralgia is presented, who underwent various interventions such as acupuncture, block therapies and even microvascular decompression without effective pain relief. Clinical findings: Paresthesias and shooting-like twinges of pain intensity 10/10 in bilateral maxillary and mandibular branches of the trigeminal nerve, with nasal and intraoral triggers that made eating impossible, becoming increasingly severe since refractoriness to microvascular decompression and carbamazepines, triggering the twinges even during sleep, generating somnolence, depressive mood and social isolation. Treatment and results: The patient was evaluated by an interdisciplinary neuro-oncology team, where, in accordance with the analysis of the brain magnetic resonance imaging and the patient's history, it was indicated to perform Cyberknife® radiosurgery in monofraction on the left trigeminal and subsequently treat the contralateral trigeminal. When treated with Cyberknife® radiosurgery, the patient reported absolute improvement in her pain for 2 years. Clinical relevance: Radiosurgery by CyberKnife is not yet the first line of management in trigeminal neuralgia, however, it should be considered since several studies have managed to demonstrate an increase in the quality of life of patients and pain relief in refractory or severe cases. of said pathology.

Thinner is a solvent that contains toluene, xylene, and other substances. The aim of this study was to examine the toxicity of thinner, xylene, and toluene on adult red flour beetles, Tribolium castaneum (Herbst), as well as the effects of thinner and toluene on gene expression. The major compounds identified in the thinner with peak area >2% were p-xylene (6%), toluene (4%), 2,4-dimethylheptane (3%), methylcyclohexane (2.75%), 2-methylheptane (2%), cyclohexanone (2.6%), and nonane (2.1%). Insects were exposed to solvents at 0-240 µl/L air for 4-48 hr. Animals that survived after a 4 hr exposure to 80 µl/L air were employed to determine mRNA expression using real-time PCR. Xylene was highly toxic, and the majority of the beetles did not survive 4 hr exposure at 40 µl/L air. The lethal concentration 50 (LC50) values for toluene, xylene and thinner at 48 hr exposure were 97.7, <40 and 99.8 µl/L air, respectively. Thinner and toluene induced lethargy and sluggish movement in treated insects, while with xylene these effects were not observed. Glutathione-S-transferase (Gst) gene expression increased after thinner and toluene exposure. Superoxide dismutase (SOD) gene expression rose after toluene exposure. These results suggest that T. castaneum provides an alternative model for the study of toxicological effects of volatile aromatic compounds.

RESUMO

Effective, ethical pest control requires the use of chemicals that are highly specific, safe, and ecofriendly. Linalool and ß-pinene occur naturally as major constituents of the essential oils of many plant species distributed throughout the world, and thus meet these requirements. These monoterpenes were tested as repellents against Tribolium castaneum, using the area preference method, after four hours of exposure and the effect transcriptional of genes associated with neurotransmission. Changes in gene expression of acetylcholinesterase (Ace1), GABA-gated anion channel splice variant 3a6a (Rdl), GABA-gated ion channel (Grd), glutamate-gated chloride channel (Glucl), and histamine-gated chloride channel 2 (Hiscl2) were assessed and the interaction with proteins important for the insect using in silico methods was also studied. For linalool and ß-pinene, the repellent concentration 50 (RC50) values were 0.11 µL/cm² and 0.03 µL/cm², respectively. Both compounds induced overexpression of Hiscl2 gen in adult insects, and ß-pinene also promoted the overexpression of Grd and the Ace1 gene. However, ß-pinene and linalool had little potential to dock on computer-generated models for GABA-gated ion channel LCCH3, nicotinic acetylcholine receptor subunits alpha1 and alpha2, and putative octopamine/tyramine receptor proteins from T. castaneum as their respective binding affinities were marginal, and therefore the repellent action probably involved mechanisms other than direct interaction with these targets. Results indicated that ß-pinene was more potent than linalool in inducing insect repellency, and also had a greater capacity to generate changes in the expression of genes involved in neuronal transmission.

RESUMO

Naphthalene and benzene are widely-used volatile organic compounds. The aim of this research was to examine the toxicological effects of naphthalene and benzene against Tribolium castaneum as an animal model. Adult insects were exposed to these aromatic compounds to assess mortality after 4-48 h of exposure. The lethal concentration 50 (LC50) for naphthalene, naphthalin, and benzene were 63.6 µL/L, 20.0 µL/L, and 115.9 µL/L in air, respectively. Real-time polymerase chain reaction (PCR) analysis revealed expression changes in genes related to oxidative stress and metabolism [Glutathione S-Transferase (Gst), and Cytochrome P450 6BQ8 (Cyp6bq8)]; reproduction and metamorphosis [Hormone receptor in 39-like protein (Hr39), Ecdysone receptor: (Ecr), and Chitin synthase 2 (Chs2)]; and neurotransmission [Histamine-gated chloride channel 2 (Hiscl2)] in insects exposed for 4 h to 70.2 µL/L naphthalene. Adults exposed to benzene (80 µL/L; 4 h) overexpressed genes related to neurotransmission [GABA-gated anion channel (Rdl), Hiscl2, and GABA-gated ion channel (Grd)]; reproduction and metamorphosis [Ultraspiracle nuclear receptor (USP), Ecr; and Hr39]; and development (Chs2). The data presented here provides evidence that naphthalene and benzene inhalation are able to induce alterations on reproduction, development, metamorphosis, oxidative stress, metabolism, neurotransmission, and death of the insect.

In this study in silico activities of essential oils components on dengue virus (DENV) were evaluated, and beta-Caryophyllene was subjected to biological examination to assess inhibition of DENV-2 replication. Components previously optimized were coupled with viral proteins prepared, using AutoDock Vina. Theoretical affinity values varied between -4.0 and -7.3 kcal/mol. alpha-copaene, beta-bourbonene, germacrene D, spathulenol, beta-caryophyllene, caryophyllene oxide and (+)- epi-bicyclosesquiphellandrene showed the greatest interaction with viral proteins. beta-caryophyllene inhibits DENV-2 in vitro (50 percent inhibitory concentration [IC50] = 22.5 +/- 5.6 uM [4.6 +/- 1.1 ug/mL] and resulted non-cytostatic with a selectivity index value of 71.1. The in silico results permit infer that DENV proteins are potential targets for the concomitant docking of various essential oils components. Biological examination suggest that beta-caryophyllene acts on very early steps of the viral replication cycle and it might prove virucidal.

Portovelo is one of the oldest gold mining towns in Ecuador. Artisanal gold mining still uses mercury in the process of gold recovery. In this study, mercury concentrations in the air of Portovelo were evaluated. High mercury levels in the ambient were found in El Pache sector, where most gold mining processing plants are located. These varied between 2,356.7 ± 1,807.6 and 3,699.5 ± 1,225.3 ng/m(3) during the rainy and dry seasons, respectively. Lower levels were detected in the urban (central) area of Portovelo, with 214.6 ± 43.7 ng/m(3) in the rainy season and 574.2 ± 72.8 ng/m(3) in the dry season, exceeding the Agency for Toxic Substances and Disease Registry minimum risk level of 200 ng/m(3). Average mercury concentrations in exhaled air from miners, measured before and after amalgam burning ranged between 179-1,352 and 2,007-3,389 ng/m(3), respectively. These data suggest Portovelo air is polluted with mercury and humans are being dangerously exposed. Therefore, strong actions must be undertaken to protect human and environmental health, including changing gold recovery systems.
